Anti-KRAS G12D mTCR PBL is an investigational cell therapy in which a patient’s own peripheral blood lymphocytes (PBLs) are harvested and genetically modified ex vivo using a retroviral vector to express a murine T-cell receptor (mTCR) that specifically recognizes the KRAS protein with the oncogenic G12D mutation, presented by HLA-A*11:01. The engineered cells are then expanded and reinfused into the patient, aiming to target and destroy cancer cells harboring this specific KRAS mutation. This approach is being developed primarily for solid tumors such as pancreatic adenocarcinoma, colorectal cancer, and other malignancies driven by KRAS G12D mutations. The therapy represents a form of adoptive T cell transfer immunotherapy designed to overcome immune tolerance to self-antigens by introducing high-affinity TCRs from mice[1][2][3][4].
