Anti-KRAS G12V murine TCR transduced PBL is an autologous T-cell receptor (TCR) engineered T-cell therapy developed by the National Cancer Institute (NCI) for the treatment of advanced solid tumors. The therapy involves the collection of a patient's peripheral blood lymphocytes (PBLs), which are then genetically modified using a retroviral vector to express a murine-derived TCR. This specific TCR is designed to recognize the KRAS G12V mutation, a common oncogenic driver, when presented by the HLA-A*11:01 MHC class I allele. The treatment protocol includes a lymphodepleting conditioning regimen of cyclophosphamide and fludarabine, followed by the intravenous infusion of the engineered T cells and the administration of aldesleukin (IL-2) to support the expansion and persistence of the cells in vivo. It is primarily being investigated for KRAS G12V-positive pancreatic, colorectal, and gastric cancers.
