Overview

Anti-L1CAM monoclonal antibodies are research-stage biologics designed to target the L1 cell adhesion molecule (L1CAM, also known as CD171), a 200-220 kDa transmembrane glycoprotein of the immunoglobulin superfamily. L1CAM is highly expressed during neural development and is aberrantly overexpressed in numerous human malignancies, where it serves as a driver of the epithelial-mesenchymal transition (EMT), tumor cell motility, and metastasis. These antibodies, such as the clone HSL175, are being investigated for their ability to disrupt L1CAM-mediated signaling and as targeting moieties for antibody-drug conjugates (ADCs). Preclinical evidence suggests that anti-L1CAM therapy can significantly reduce perineural invasion (PNI) and inhibit the growth of small cell lung cancer (SCLC), particularly the NEUROD1-dominant subtype, as well as head and neck cancers.
