The anti-LAG-3 IgG4 antibody is a monoclonal antibody designed to target the Lymphocyte Activation Gene-3 (LAG-3), an inhibitory checkpoint receptor expressed on the surface of activated T cells. LAG-3 negatively regulates T-cell activation and effector functions upon binding to its ligands, such as MHC class II molecules, contributing to T-cell exhaustion in the tumor microenvironment. By blocking this interaction, the antibody aims to restore T-cell activity and enhance anti-tumor immune responses. The IgG4 isotype is typically selected for these therapeutic antibodies to provide potent pathway blockade while minimizing effector functions like antibody-dependent cellular cytotoxicity (ADCC). In preclinical research, such as studies conducted by MacroGenics, this antibody has been shown to reverse mesenchymal stromal cell (MSC)-mediated immunosuppression in the acute myeloid leukemia (AML) microenvironment, leading to increased T-cell-mediated killing of AML cells.
