anti-LAIR1 antibody is a monoclonal antibody designed to target and block the Leukocyte-associated Ig-like receptor 1 (LAIR1), also known as CD305. LAIR1 is an inhibitory immune receptor predominantly expressed on myeloid lineage cells, specifically M2-like tumor-associated macrophages (TAMs), which are abundant in the tumor microenvironment (TME) of aggressive cancers. The antibody functions by preventing the interaction between LAIR1 and its ligands, such as collagen IV, thereby disrupting an immunosuppressive signaling pathway. This blockade promotes the repolarization of macrophages from a pro-tumor M2-like phenotype to an anti-tumor M1-like phenotype, reduces the secretion of Factor XIII-A (FXIII-A), and decreases collagen IV deposition. Developed by researchers at the University of Florida, the antibody is intended to enhance T-cell infiltration and activity, offering a potential therapeutic strategy for "cold" tumors like glioblastoma and lung cancer that are often resistant to standard checkpoint inhibitors like PD-1 blockades.
