Anti-LAMP5 ADC-MMAE is a preclinical antibody-drug conjugate (ADC) that targets Lysosomal Associated Membrane Protein Family Member 5 (LAMP5), a protein found exposed on the plasma membrane of malignant plasma cells in multiple myeloma. The conjugate consists of a monoclonal antibody directed against the extracellular domain of LAMP5, linked to the potent antimitotic cytotoxic payload monomethyl auristatin E (MMAE). In preclinical studies, the ADC has demonstrated the ability to reduce the viability of LAMP5-positive myeloma cells in a time- and concentration-dependent manner, activate immunogenic cell death in vitro, and exhibit anti-tumor activity in vivo. It is being investigated as a potential therapeutic approach for high-risk multiple myeloma, particularly in patients where high LAMP5 expression correlates with poor prognosis and increased disease burden. The asset was developed by researchers at the University of Texas MD Anderson Cancer Center.
