Anti-LAMP5 ADC-PBD is a preclinical antibody-drug conjugate (ADC) designed to target Lysosomal Associated Membrane Protein Family Member 5 (LAMP5), which is overexpressed on the surface of malignant plasma cells in high-risk multiple myeloma. The conjugate consists of a monoclonal antibody specific to the extracellular domain of LAMP5 linked to a pyrrolobenzodiazepine (PBD) dimer cytotoxic payload. Upon binding and internalization, the PBD dimer induces DNA cross-linking, leading to cell death. Research presented at ASH 2023 demonstrated that this ADC reduces the viability of LAMP5-positive myeloma cells, activates immunogenic cell death, and exhibits anti-tumor activity in vivo.
