anti-LAT1 shRNA is a short hairpin RNA designed to silence the expression of the Large Neutral Amino Acid Transporter 1 (LAT1), also known as SLC7A5. LAT1 is a transmembrane protein that facilitates the transport of large neutral amino acids and is frequently overexpressed in various cancers, including glioblastoma, where it supports rapid tumor growth and metabolic demands. In research settings, such as studies conducted at the Mayo Clinic, anti-LAT1 shRNA is typically delivered via lentiviral vectors to human glioblastoma cell lines and xenografts. The knockdown of LAT1 results in a significant reduction in the uptake of amino acid analogs like 3H-L-DOPA (a surrogate for the PET tracer 18F-FDOPA), confirming LAT1's role as a primary transporter for these imaging agents and a potential therapeutic target.
