Overview

An anti-leptin monoclonal antibody is a therapeutic agent designed to target and neutralize leptin, a peptide hormone primarily secreted by adipocytes that regulates energy balance, food intake, and body weight. While leptin was initially studied as an anti-obesity hormone, individuals with obesity often develop hyperleptinemia and leptin resistance. Research, particularly led by UT Southwestern and UTHealth, has shown that partial reduction of circulating leptin using a neutralizing monoclonal antibody (such as hLep3 or LepAb) can restore leptin sensitivity, reduce body weight, and improve glucose tolerance. Additionally, anti-leptin monoclonal antibodies are being investigated for their potential to overcome treatment resistance in estrogen receptor-positive (ER+) breast cancer (by targeting the obesity-breast cancer axis) and to mitigate tissue fibrosis in the liver, kidney, and lungs.
