The anti-Lewis Y antibody is a protease-activated pro-antibody being co-developed by PrecisemAb Biotech and GlycoNex for the treatment of various cancers. It targets the Lewis Y (LeY) antigen, a carbohydrate glycan that is overexpressed on the surface of many epithelial tumors, including those of the breast, lung, colon, and ovary. The drug utilizes PrecisemAb's proprietary Universal Antibody Masking technology, which involves an inhibitory masking peptide linked to the antibody via a protease-cleavable sequence. This design ensures the antibody remains inactive in healthy tissues, reducing systemic toxicity, and is only activated within the tumor microenvironment where specific proteases are present to cleave the mask. Once activated, the antibody binds to Lewis Y on cancer cells to mediate anti-tumor effects, potentially through antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC).
