anti-LGALS3BP ADC is an antibody-drug conjugate (ADC) being developed by MediaPharma for the treatment of various solid tumors, including neuroblastoma, glioblastoma, and adenoid cystic carcinoma. The therapeutic consists of a humanized monoclonal antibody (derived from the 1959 clone) that specifically targets Galectin-3 Binding Protein (LGALS3BP, also known as 90K), a secreted glycoprotein that is highly expressed on the surface of cancer cells and within the tumor microenvironment. The antibody is conjugated to a cytotoxic payload, typically monomethyl auristatin F (MMAF), via a linker. Upon binding to LGALS3BP, the ADC is internalized through endocytosis, leading to the lysosomal degradation of the complex and the subsequent release of the payload. The released payload inhibits tubulin polymerization, resulting in cell cycle arrest and apoptosis of the target cancer cells. The program is currently in preclinical development.
