anti-LRP6 refers to a class of monoclonal antibodies designed to target the Low-density lipoprotein receptor-related protein 6 (LRP6), a critical co-receptor in the canonical Wnt/β-catenin signaling pathway. While many LRP6-targeted antibodies are developed as antagonists to inhibit Wnt-driven tumor growth, specific agents developed by Novartis act as Wnt signaling potentiators. These antibodies bind to LRP6 and enhance Wnt-induced signal transduction, which is essential for osteoblast differentiation and bone formation. In preclinical models of multiple myeloma, anti-LRP6 therapy has demonstrated the ability to prevent myeloma-induced bone loss by promoting bone anabolic activity and reducing bone resorption. This therapeutic strategy is often explored in combination with other Wnt-modulating agents, such as DKK1 inhibitors, to restore skeletal integrity in patients with osteolytic bone disease.
