Anti-Ly6G is a monoclonal antibody that specifically binds to the Ly6G antigen expressed on mouse neutrophils. By recognizing the Ly6G cell surface protein, anti-Ly6G induces depletion of Ly6G-positive neutrophils primarily through Fc-dependent mechanisms, notably macrophage-mediated phagocytosis. The antibody is used extensively in mouse research to study neutrophil function in various pathological contexts, including tumor biology, autoimmune disease, and infection. While anti-Ly6G is considered specific for neutrophil depletion (in contrast to anti-Gr1, which also recognizes Ly6C), depletion is incomplete due to the emergence of Ly6G^low^ neutrophils resistant to antibody-mediated clearance. Anti-Ly6G treatment also primes residual neutrophils for increased oxidative burst upon inflammatory stimulation and can induce anti-neutrophil cytoplasmic autoantibodies (ANCA) in vivo. The antibody’s mechanism includes Ly6G internalization and masking, which may impede consistent detection by subsequent antibodies or flow cytometry[1][3][4][5][8][9].
