Anti-Ly6G neutralizing antibody is a monoclonal antibody used primarily in preclinical research to selectively deplete neutrophils in murine models. It targets Ly6G, a 21-25 kDa glycosylphosphatidylinositol (GPI)-anchored protein that is a specific marker for mature murine neutrophils. Unlike anti-Gr-1 antibodies (such as clone RB6-8C5), which target both Ly6G and Ly6C and thus deplete both neutrophils and monocytes, anti-Ly6G antibodies (most notably clone 1A8) are highly specific for neutrophils. In oncology research, these antibodies are employed to investigate the role of neutrophils in the tumor microenvironment, where they are often associated with immune tolerance and tumor rebound following chemotherapy. Depletion is typically achieved via intraperitoneal administration, leading to a rapid reduction in circulating and tissue-resident neutrophils through Fc-mediated mechanisms such as antibody-dependent cellular cytotoxicity (ADCC).
