Overview

Anti-lymphocyte serum (ALS) produced by Hospices Civils de Lyon is a rabbit-derived polyclonal antibody preparation used primarily as an immunosuppressive agent in the context of allogeneic hematopoietic stem cell transplantation (HSCT). It is specifically employed in conditioning regimens to prevent graft-versus-host disease (GvHD) and to facilitate engraftment by depleting both host and donor T-lymphocytes. The serum contains a diverse array of antibodies targeting various T-cell surface antigens, including CD2, CD3, CD4, CD8, and HLA molecules. Its mechanism of action involves the depletion of T-cells through complement-dependent cytotoxicity (CDC), antibody-dependent cellular cytotoxicity (ADCC), and the induction of apoptosis. Developed and manufactured by the Hospices Civils de Lyon, this specific ALS formulation is a cornerstone of the "Lyon regimen" and has been evaluated in numerous clinical trials to optimize transplant outcomes.
