Overview

Anti-MAGE-A3-DP4 T cell receptor peripheral blood lymphocytes is an investigational autologous cell therapy in which a patient’s own CD4+ peripheral blood lymphocytes are genetically engineered to express a high-affinity, HLA-DPB1*04:01 or *04:02-restricted T cell receptor (TCR) that specifically targets the cancer-testis antigen MAGE-A3. The modified cells are expanded ex vivo and infused back into the patient following a nonmyeloablative, lymphodepleting conditioning regimen. This approach aims to direct the immune system to recognize and attack tumor cells expressing MAGE-A3 (and closely related MAGE-A6), which are overexpressed in various solid tumors but not in most normal tissues except testes. The therapy has shown evidence of clinical activity—including complete and partial responses—in patients with metastatic cancers such as cervical cancer, esophageal cancer, urothelial carcinoma, and osteosarcoma. It is being developed primarily by the National Cancer Institute (NCI) with support from Kite for next-generation manufacturing[2][3][7].
