Anti-MARCO antibody is a **monoclonal antibody** targeting the macrophage receptor with collagenous structure (MARCO), a member of the class A scavenger receptor family found on distinct populations of macrophages, especially tumor-associated macrophages (TAMs)[1][3][5]. The antibody binds to the MARCO receptor and induces its internalization, reprograms the metabolic and phenotypic profile of MARCO-expressing TAMs, and activates natural killer (NK) cells via enhanced IL-15 production and TRAIL-dependent tumor cell killing[1][12]. By modifying the tumor microenvironment, anti-MARCO antibody reduces immunosuppression, increases inflammatory signals, and synergizes with immunotherapies such as anti-PD1, anti–PD-L1, and anti-CTLA4 to promote tumor regression, especially in melanoma and other solid tumors[1][2][4][10][12]. This therapeutic strategy is also being translated to human application, with preclinical and phase 1 clinical investigations ongoing in advanced solid tumors[8][9][12].
