Drug intelligence / Profile preview
An anti-MARCO blocking antibody is a monoclonal antibody designed to target and inhibit Macrophage Receptor with Collagenous Structure (MARCO), a class A scavenger receptor primarily expressed on immunosuppressive tumor-associated macrophages (TAMs) and tissue-resident macrophages. In oncology, blocking MARCO reprogrammes TAMs from an immunosuppressive M2-like phenotype to a pro-inflammatory M1-like phenotype, enhancing anti-tumor immunity and improving the efficacy of immune checkpoint inhibitors. Additionally, preclinical research has identified MARCO as an alternative entry receptor for SARS-CoV-2 in macrophages, where it facilitates viral entry and subsequent pyroptosis. Consequently, anti-MARCO blocking antibodies are being investigated as potential therapeutics to reduce the severity of COVID-19 and treat various solid tumors.
