Overview

The anti-MART-1 F5 T-cell receptor is an engineered, high-affinity, HLA-A*0201-restricted human T cell receptor (TCR) specific for the melanoma antigen recognized by T cells 1 (MART‑1), also known as Melan-A. This therapy involves isolating a patient’s own peripheral blood lymphocytes (PBLs), genetically modifying them ex vivo using a retroviral vector to express the alpha and beta chains of the DMF5 clone of the MART‑1-specific TCR, expanding these modified cells, and then reinfusing them into the patient after a non-myeloablative conditioning regimen. The engineered cells are designed to recognize and kill melanoma cells expressing MART‑1. This approach is being developed primarily for patients with advanced or metastatic melanoma who are HLA-A*0201-positive[3][4][6][8]. The therapy has been tested in phase 2 clinical trials in combination with dendritic cell vaccination and interleukin 2 administration[3][6]. The main mechanism is targeted immunotherapy via adoptive transfer of tumor-reactive cytotoxic lymphocytes.
