The anti-mesothelin × cadherin-3 bispecific antibody-drug conjugate is an experimental therapeutic developed by Biocytogen for the treatment of solid tumors, specifically ovarian and pancreatic cancers. Utilizing the proprietary RenLite® common light chain mouse platform, the drug consists of a fully human bispecific antibody with a monovalent '1+1' architecture that simultaneously targets Mesothelin (MSLN) and Cadherin-3 (CDH3/P-cadherin). This dual-targeting approach is designed to improve tumor specificity, enhance cellular internalization, and overcome potential antigen escape mechanisms in tumors where these two antigens are co-expressed or spatially complementary. The antibody is conjugated to cytotoxic payloads, such as the microtubule inhibitor monomethyl auristatin E (MMAE) or a novel DNA topoisomerase inhibitor payload, BLD1102. Preclinical data indicate that this bispecific ADC (bsADC) exhibits potent antigen-dependent cytotoxicity and superior anti-tumor activity in patient-derived xenograft models compared to its monospecific counterparts.
