Overview

Anti-mesothelin-7×19 CAR-T cells are autologous chimeric antigen receptor (CAR)-T cells genetically engineered to express a CAR targeting mesothelin (MSLN), a tumor-associated antigen frequently overexpressed on various solid tumors. These T cells are further modified to secrete interleukin-7 (IL-7) and CCL19 (7×19), two factors intended to enhance T-cell migration, infiltration, and persistence within the tumor microenvironment. The IL-7 and CCL19 are linked via a 2A peptide and co-expressed using a dual lentiviral vector system. Preclinical and early clinical studies indicate superior expansion, migration, and antitumor activity compared to conventional anti-mesothelin CAR-T cells, especially in pancreatic carcinoma, hepatocellular carcinoma, and ovarian carcinoma. A phase 1 clinical trial has been conducted (e.g., NCT03198546) with promising responses noted in solid tumor patients expressing mesothelin[1].
