Overview

Anti-mesothelin CAR T-cells (NCI) is an experimental autologous cell therapy developed by the National Cancer Institute's Surgery Branch for the treatment of mesothelin-expressing metastatic cancers. The therapy involves the ex vivo genetic modification of a patient's peripheral blood lymphocytes using a retroviral vector to express a chimeric antigen receptor (CAR) specific for mesothelin, a cell-surface glycoprotein frequently overexpressed in mesothelioma, pancreatic, ovarian, and lung cancers. The treatment regimen includes lymphodepleting conditioning with cyclophosphamide and fludarabine to create a favorable environment for T-cell expansion, followed by the infusion of the engineered cells and subsequent administration of high-dose aldesleukin (IL-2) to enhance the persistence and anti-tumor activity of the CAR T-cells.
