Overview

Anti-mesothelin nanobody chimeric antigen receptor natural killer (CAR-NK) cells are an investigational cell therapy designed to target mesothelin-expressing solid tumors. This therapy utilizes a nanobody (a single-domain VHH antibody fragment) as the antigen-binding domain of the CAR, which offers advantages such as a smaller molecular size, high stability, and the ability to easily construct tandem CARs to address tumor heterogeneity. These CAR-NK cells are typically derived from umbilical cord blood and engineered via third-generation lentiviral transduction to express the CAR construct. Upon administration, the cells recognize and bind to mesothelin (MSLN) on the surface of cancer cells, triggering NK cell activation and potent cytotoxicity. Preclinical studies have demonstrated significant efficacy in suppressing tumor growth in xenograft models of pancreatic cancer, highlighting the potential of nanobody-based designs for targeting cell-surface antigens in solid malignancies.
