Anti-MICB CAR-NK is an investigational chimeric antigen receptor (CAR)-modified natural killer (NK) cell therapy designed to target MHC class I polypeptide-related sequence B (MICB), a stress-induced ligand frequently overexpressed on various solid tumors, including pancreatic cancer. The construct typically incorporates anti-MICB single-chain variable fragments (scFvs) for tumor recognition, coupled with a CD8 hinge and transmembrane domain, a 4-1BB co-stimulatory domain, and a CD3ζ activation domain. A unique feature of this specific design is the inclusion of an IL-15 expression cassette, which promotes NK cell persistence and activation, and the secretion of free anti-MICB scFvs to block tumor immune evasion mediated by soluble MICB. By targeting MICB, these CAR-NK cells induce direct cytotoxicity through the release of perforin and granzymes and the secretion of pro-inflammatory cytokines like IFN-γ and TNF-α.
