Overview

Anti-Mincle antibodies are therapeutic agents designed to bind and neutralize the Macrophage-inducible C-type lectin (Mincle), a member of the C-type lectin receptor (CLR) family encoded by the CLEC4E gene. Mincle is primarily expressed on myeloid cells, such as macrophages and neutrophils, and functions as a pattern recognition receptor that detects both pathogen-associated molecular patterns (PAMPs) and damage-associated molecular patterns (DAMPs), such as SAP130 released from necrotic cells. Upon activation, Mincle signals through the FcRγ-Syk-CARD9 pathway to induce the production of pro-inflammatory cytokines and chemokines. In the context of Crohn's disease and other inflammatory conditions, Mincle signaling has been shown to promote intestinal mucosal inflammation, macrophage pyroptosis, and neutrophil chemotaxis. Neutralizing antibodies against Mincle are being investigated in preclinical models to ameliorate inflammation by blocking these pathways, suggesting a potential therapeutic role in treating autoimmune and inflammatory disorders.
