anti-miR-146a refers to a class of antisense oligonucleotides (ASOs) or antagomirs designed to specifically bind and inhibit the function of microRNA-146a (miR-146a). miR-146a is a small non-coding RNA that acts as a critical regulator of the innate immune response and inflammatory signaling pathways. In the context of oncology, particularly melanoma brain metastasis, miR-146a is often upregulated and delivered via tumor-derived exosomes to the brain microenvironment. Once internalized by astrocytes, miR-146a triggers their activation, promoting the establishment of a pro-metastatic niche that facilitates tumor growth and survival. By sequestering miR-146a, anti-miR-146a agents can reverse astrocyte activation, reduce the production of inflammatory cytokines, and inhibit the development of brain metastases. Research into anti-miR-146a is primarily in the preclinical stage, exploring its potential in treating both metastatic cancers and various inflammatory or autoimmune conditions.
