Overview

anti-miR-196b antagomir is a synthetic, chemically modified antisense oligonucleotide designed to specifically inhibit the function of microRNA-196b (miR-196b). miR-196b is recognized as an oncogenic microRNA (oncomiR) that is significantly upregulated in several cancers, including MLL-rearranged acute myeloid leukemia (AML), glioblastoma, and colorectal cancer. In these contexts, miR-196b promotes oncogenesis by downregulating tumor suppressor genes such as Fas, HOXA9, and MEIS1. The antagomir chemistry typically involves 2-O-methyl sugar modifications, phosphorothioate linkages, and a 3-cholesterol tail, which collectively enhance metabolic stability, binding affinity, and cellular uptake without the need for specialized delivery vehicles. By competitively binding to the mature miR-196b sequence, the antagomir prevents its incorporation into the RNA-induced silencing complex (RISC), thereby restoring the expression of target genes and inducing anti-tumor effects. While primarily utilized as a high-potency research tool for functional genomics, it represents a therapeutic strategy for microRNA-driven malignancies.
