Anti-miR-21 PNA is a research-stage antisense oligonucleotide construct utilizing peptide nucleic acid (PNA) technology to target and silence microRNA-21 (miR-21). miR-21 is an oncomiR frequently overexpressed in various malignancies, including glioblastoma and lung cancer, where it promotes cell survival, invasion, and radioresistance by downregulating tumor suppressors like PTEN and activating the PI3K-Akt signaling pathway. In PNAs, the sugar-phosphate backbone is replaced by a neutral N-(2-aminoethyl) glycine backbone, providing high enzymatic stability and strong binding affinity to target RNA. Preclinical studies have explored various delivery methods, such as pHLIP peptide conjugation for tumor targeting or nanoparticle encapsulation, demonstrating efficacy in reducing tumor growth and reversing cardiac fibrosis.
