Drug intelligence / Profile preview
Anti-miR-34a (also known as miR-34a inhibitor or antimiR-34a) is a class of synthetic antisense oligonucleotides designed to target and inhibit microRNA-34a (miR-34a). MiR-34a is a key post-transcriptional regulator that is aberrantly elevated in metabolic disorders, obesity, and cardiovascular diseases. By binding complementarily to mature miR-34a, anti-miR-34a prevents it from suppressing its target genes, such as SIRT1, NAMPT, and beta-klotho (KLB). In preclinical models of obesity and metabolic dysfunction-associated steatohepatitis (MASH), inhibition of miR-34a restores NAD+ levels, enhances SIRT1 activity, improves FGF19/FGF21 signaling, and alleviates hepatic steatosis, inflammation, and glucose intolerance. Additionally, anti-miR-34a has been investigated for its potential to attenuate cardiac remodeling, hypertrophy, and fibrosis in cardiovascular disease models.
