Anti-miR21-conjugated DNA nanohydrogel is an experimental gene therapy platform designed for targeted cancer treatment. It consists of a biocompatible and physiologically stable DNA hydrogel that is functionalized with antisense oligonucleotides targeting microRNA-21 (miR-21), an oncogenic miRNA upregulated in various cancers such as glioblastoma, breast cancer, colon cancer, and pancreatic cancer. The mechanism involves the delivery of anti-miR-21 to tumor cells where it binds and inhibits miR-21 activity. This inhibition leads to the upregulation of tumor suppressor genes like PTEN and PDCD4, resulting in reduced cell proliferation and increased apoptosis. The hydrogel can also serve as a drug delivery vehicle by intercalating chemotherapeutic agents (e.g., doxorubicin), enabling combination chemo/gene therapy with synergistic anticancer effects[1][6]. This approach addresses challenges associated with the instability and poor cellular uptake of naked antisense oligonucleotides.
