Overview

4H2 is a monoclonal antibody specifically designed to target and block the mouse programmed cell death protein 1 (PD-1) receptor. It serves as a preclinical surrogate for human anti-PD-1 checkpoint inhibitors, such as nivolumab, allowing researchers to study the effects of PD-1 blockade in syngeneic mouse tumor models. By binding to the PD-1 receptor on T cells, 4H2 prevents interaction with its ligands, PD-L1 and PD-L2, thereby reversing T-cell exhaustion and enhancing the immune system's ability to recognize and destroy cancer cells. In preclinical studies, 4H2 is frequently used in combination with other immunotherapies, such as anti-CTLA-4 antibodies, to demonstrate synergistic anti-tumor activity and favorable changes in the tumor microenvironment, including increased effector T-cell infiltration.
