**anti-MSLN-7×19 CAR-T** is a chimeric antigen receptor T cell therapy engineered to target mesothelin (MSLN) on solid tumors. The "7×19" refers to the secretion of interleukin-7 (IL-7) and CCL19 chemokine by the CAR-T cells, which enhances their migration, infiltration, and survival within the tumor microenvironment, boosting antitumor activity. The CAR construct combines a targeting domain for mesothelin with intracellular signaling domains (CD28, TLR2). Preclinical studies and a phase 1 clinical trial have demonstrated increased efficacy against MSLN-positive pancreatic carcinoma, hepatocellular carcinoma, and ovarian carcinoma. Administration of these engineered cells resulted in complete or partial responses in advanced cancer patients, with minimal severe toxicity reported[1].
