The anti-mutated CALR immune therapeutic is a biological agent, specifically a monoclonal antibody, developed by MyeloPro in collaboration with Janssen. It is designed to selectively target the C-terminal neoepitope of mutated calreticulin (CALR), a driver mutation found in a significant proportion of patients with myeloproliferative neoplasms (MPNs), including essential thrombocythemia (ET) and primary myelofibrosis (PMF). In these diseases, the CALR mutation creates a novel protein sequence that constitutively binds to and activates the thrombopoietin receptor (MPL), leading to uncontrolled cell proliferation. This therapeutic binds to the mutant CALR protein on the cell surface to inhibit oncogenic signaling and induce immune-mediated clearance of the malignant clones. It is currently being investigated in Phase 1 clinical trials to evaluate its safety and efficacy in patients with CALR-mutant MPNs.
