Anti-NDC80 TCRm CAR-T is an investigational T-cell receptor mimic (TCRm) chimeric antigen receptor (CAR) T-cell therapy developed by Eureka Therapeutics in collaboration with Memorial Sloan Kettering Cancer Center. Utilizing Eureka's proprietary E-ALPHA platform, the therapy features a TCR mimic single-chain variable fragment (scFv), specifically the NDC80-clC clone, that recognizes the ALNEQIARL peptide derived from the intracellular NDC80 (kinetochore-associated) protein. This peptide is presented on the cell surface in the context of the HLA-A*02 complex. Unlike traditional CAR-T therapies that target surface-expressed proteins, this TCRm approach allows for the targeting of intracellular antigens typically only accessible to T-cell receptors. Preclinical studies have demonstrated high specificity and potent anti-tumor activity against various HLA-A*02-positive malignancies, including acute lymphoblastic leukemia (ALL), acute myeloid leukemia (AML), and several solid tumors, while sparing healthy hematopoietic stem cells and leukocytes.
