The anti-nectin-4 parental antibody-drug conjugate is a monovalent ADC targeting Nectin-4 (Poliovirus receptor-related protein 4), developed by Biocytogen as a foundational component and benchmark for their bispecific ADC program, BCG039 (a Nectin-4 x TROP2 bispecific ADC). The antibody component is a fully human monoclonal antibody generated using Biocytogen's RenLite transgenic mouse platform. In preclinical studies, this parental ADC has been conjugated to various payloads, including the microtubule inhibitor monomethyl auristatin E (MMAE) via a valine-citrulline (vc) linker and Biocytogen's proprietary BLD1102 linker-payload, which features a novel topoisomerase 1 (TOP1) inhibitor. It is used in comparative assays to demonstrate the superior internalization and anti-tumor efficacy of the bispecific format in models of urothelial carcinoma and other solid tumors.
