The anti-NEDD9 antibody is a novel therapeutic candidate designed to inhibit platelet–pulmonary endothelial adhesion and prevent chronic thromboembolic vascular remodeling. Developed by researchers at institutions including Brigham and Women’s Hospital and Massachusetts General Hospital, the antibody specifically targets NEDD9 (Neural precursor cell expressed, developmentally down-regulated 9), a hypoxia-sensitive protein that is upregulated on the plasma membrane of pulmonary artery endothelial cells. By blocking the protein-protein interaction between endothelial NEDD9 and the platelet surface adhesion molecule P-selectin, the antibody inhibits the adhesion of activated platelets to the pulmonary circulation. Preclinical studies in animal models of chronic thromboembolic pulmonary hypertension (CTEPH) and pulmonary hypertension have demonstrated that treatment with the anti-NEDD9 antibody reduces platelet-endothelial aggregates and attenuates vascular remodeling, highlighting its potential as a targeted therapy for disorders involving increased hypoxia signaling.
