Overview

Anti-nicastrin monoclonal antibody is a research-stage biologic therapeutic agent targeting nicastrin (NCSTN), an essential component of the gamma-secretase complex. Developed primarily at Imperial College London, this monoclonal antibody blocks nicastrin on the cell surface, thereby inhibiting gamma-secretase enzymatic activity and downstream oncogenic Notch signaling. It has been investigated in preclinical studies for the treatment of invasive breast cancers, particularly triple-negative breast cancer (TNBC), as well as other solid tumors and hematological malignancies. Specific clones, such as 2H6 and 10C11, have demonstrated the ability to reduce cancer cell proliferation, invasion, and epithelial-to-mesenchymal transition (EMT) in vitro and in vivo.
