This preclinical biologic, developed through a collaboration between the Walter and Eliza Hall Institute (WEHI) and Roche (Genentech), is a monoclonal antibody designed to block the protein Ninjurin-1 (NINJ1). NINJ1 is a critical mediator of plasma membrane rupture (PMR), which represents the final, irreversible stage of various forms of programmed lytic cell death, including pyroptosis, necroptosis, and certain types of apoptosis. By binding to NINJ1 and preventing its oligomerization into membrane-disrupting pores, the antibody inhibits the explosive release of intracellular contents, such as pro-inflammatory cytokines (e.g., IL-1β) and damage-associated molecular patterns (DAMPs). This blockade aims to mitigate the systemic inflammation and tissue damage associated with various cell death-driven pathologies, including sepsis, ischemia-reperfusion injury, and autoimmune diseases.
