Drug intelligence / Profile preview
An anti-NKG2D monoclonal antibody is a targeted biologic therapy designed to bind to the Natural Killer Group 2 Member D (NKG2D) receptor (also known as CD314 or KLRK1). NKG2D is an activating receptor expressed on natural killer (NK) cells, CD8+ T cells, and other T cell subsets. By binding to NKG2D, these monoclonal antibodies block its interaction with stress-induced ligands (such as MICA, MICB, and ULBP proteins), thereby inhibiting the activation, proliferation, and cytotoxic activity of NKG2D-expressing immune cells. This therapeutic approach is primarily investigated for the treatment of autoimmune and inflammatory diseases, such as Crohn's disease and rheumatoid arthritis, where aberrant NKG2D signaling drives pathological tissue damage, as well as in transplant rejection models to prevent acute antibody-mediated rejection.
