An anti-NKG2D neutralizing antibody is a monoclonal antibody designed to bind to the NKG2D receptor (Natural Killer Group 2, Member D; also known as KLRK1) and block its interaction with its various ligands, including MICA, MICB, and the ULBP family (ULBP1-6). NKG2D is a major activating receptor expressed on natural killer (NK) cells, CD8+ T cells, and certain subsets of γδ T cells. By neutralizing this receptor, these antibodies inhibit the activation and cytotoxic activity of these immune cells. In oncology research, such as the study of glioblastoma stem cells (GSCs), these antibodies are utilized to investigate immune evasion mechanisms, as GSCs often downregulate NKG2D ligands to escape NK cell-mediated lysis. Clinically, NKG2D blockade has been explored as a therapeutic strategy for autoimmune and inflammatory conditions, such as Crohn's disease and rheumatoid arthritis, where aberrant NKG2D signaling contributes to pathological tissue damage.
