An anti-NKp46 monoclonal antibody is a type of immunotherapy designed to target and bind to NKp46 (also known as NCR1 or CD335), a key activating receptor expressed specifically on natural killer (NK) cells. NKp46 is a member of the natural cytotoxicity receptor (NCR) family and plays a critical role in the recognition and lysis of tumor cells and virus-infected cells. By binding to NKp46, these antibodies can trigger NK cell activation, degranulation, and the production of pro-inflammatory cytokines such as IFN-gamma and TNF-alpha. In the context of cancer therapy, anti-NKp46 antibodies are often utilized as the NK-cell-engaging arm of multi-specific antibodies (e.g., bispecific or trispecific NK cell engagers), which simultaneously bind to a tumor-associated antigen and NKp46 to redirect NK cell cytotoxicity toward the cancer cells. Standalone anti-NKp46 antibodies have also demonstrated the ability to induce NK cell-mediated killing of various malignancies, including multiple myeloma, in preclinical models.
