Overview

Anti-NPM1c CAR-T cells are an experimental adoptive cellular immunotherapy designed to target a neoepitope derived from the mutated nucleophosmin (NPM1c) protein, which is a common driver mutation in approximately 35% of acute myeloid leukemia (AML) cases. This specific CAR-T construct utilizes a human single-chain variable fragment (scFv) that recognizes the NPM1c peptide presented by the HLA-A2 MHC class I molecule. By targeting an intracellular protein-derived neoantigen presented on the cell surface, this therapy aims to achieve high tumor specificity and minimize on-target/off-tumor toxicities associated with targeting broadly expressed surface antigens. Preclinical studies conducted by researchers at the Dana-Farber Cancer Institute and MIT have demonstrated that these CAR-T cells exhibit potent cytotoxicity against NPM1c-positive, HLA-A2-positive AML cell lines and primary patient blasts in vitro and in vivo.
