This investigational cell therapy consists of autologous peripheral blood lymphocytes (PBLs) that have been genetically modified using a retroviral vector to express a murine T-cell receptor (mTCR) specific for the NY-ESO-1 cancer-testis antigen. Developed by the Albert Einstein College of Medicine, the therapy is designed for HLA-A2+ patients with metastatic malignancies that express NY-ESO-1. The treatment involves harvesting the patient's own lymphocytes, transducing them ex vivo with the mTCR, and reinfusing them following lymphodepleting chemotherapy (typically cyclophosphamide and fludarabine). The therapy is administered in combination with aldesleukin (IL-2) to enhance the expansion and persistence of the engineered T cells, which are programmed to recognize and eliminate tumor cells expressing the NY-ESO-1 antigen.
