Anti-NY ESO-1 TCR-transduced T cells is an autologous adoptive T-cell therapy developed by Shenzhen Second People's Hospital. The therapy involves isolating peripheral blood mononuclear cells (PBMCs) from a patient and engineering them using a lentiviral or retroviral vector to express a T-cell receptor (TCR) specific for the NY-ESO-1 (New York Esophageal Squamous Cell Carcinoma-1) cancer-testis antigen. Specifically, the TCR is HLA-A2-restricted and targets the NY-ESO-1:157-165 epitope. After in vitro expansion, the engineered T cells are infused back into the patient following a lymphodepleting preconditioning regimen. This investigational biologic is being evaluated in Phase I clinical trials for various HLA-A2-positive, NY-ESO-1-expressing relapsed or refractory malignancies, including synovial sarcoma, melanoma, and multiple myeloma.
