Anti-O-acetylated-GD2 monoclonal antibody is an investigational immunotherapy designed to target O-acetylated-GD2 (OAcGD2) ganglioside, an antigen found in high concentrations on the surface of various cancer cells, including neuroblastoma, glioblastoma, and breast cancer stem cells. Unlike the non-acetylated GD2, OAcGD2 is not significantly expressed on normal peripheral nerve fibers, which aims to mitigate the severe neuropathic pain commonly associated with anti-GD2 therapies. The antibody exerts its anti-tumor effects through multiple mechanisms, including antibody-dependent cellular cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), and direct induction of tumor cell apoptosis via pathways involving caspase-3. Developed by OGD2 Pharma, this drug has received orphan drug designation for neuroblastoma and is currently in preclinical development for several oncology indications.
