Overview

The anti-P329G adapter CAR T-cell is a modular chimeric antigen receptor (CAR) T-cell therapy platform designed to overcome limitations of traditional CAR T-cells in solid tumors, such as antigen heterogeneity and on-target/off-tumor toxicity. This system separates the antigen recognition component from the CAR signaling component. The CAR T-cells are engineered to recognize a specific P329G point mutation in the Fc region of human IgG1 antibodies. By administering "adapter" antibodies (e.g., P329G-Fc-mutated antibodies targeting EGFR), the CAR T-cells can be precisely and reversibly redirected toward tumor cells. This approach allows for tunable activity and the ability to target multiple antigens sequentially or simultaneously. Research conducted by LMU Munich and Roche has demonstrated efficacy in lung cancer models, specifically targeting EGFR while sparing non-malignant tissue.
