Anti-P329G CAR T cells are a modular, universal chimeric antigen receptor (CAR) T cell platform designed to overcome the limitations of conventional CAR T therapies. Instead of directly targeting a tumor-associated antigen, these engineered T cells express a CAR that specifically recognizes the P329G mutation within the Fc region of IgG1 antibodies (specifically those carrying the P329G L234A/L235A, or PGLALA, Fc-silencing mutations). This design makes the CAR T cells dependent on the co-administration of P329G-mutated "adaptor" antibodies to bridge the T cells to the tumor cells. By switching the adaptor antibody, the same CAR T cell product can be redirected against various antigens, including CD20, HER2, FOLR1, EpCAM, FAP, CD33, CD123, and CSF1R. This platform was developed by researchers at Roche and Genentech and is currently in the preclinical research stage for the treatment of B-cell malignancies and acute myeloid leukemia (AML).
