Anti-p40 refers to a class of monoclonal antibodies that target the p40 subunit (encoded by the IL12B gene), which is a shared component of the heterodimeric cytokines interleukin-12 (IL-12) and interleukin-23 (IL-23). By binding to the p40 subunit, these antibodies prevent IL-12 and IL-23 from interacting with their respective receptor complexes, thereby inhibiting the activation of Th1 and Th17 immune responses. This mechanism is a cornerstone in the treatment of various immune-mediated inflammatory diseases. In clinical practice, ustekinumab (Stelara) is the primary approved anti-p40 therapy, used for conditions such as plaque psoriasis, psoriatic arthritis, Crohn's disease, and ulcerative colitis. In preclinical research, anti-p40 often refers to surrogate antibodies used to validate the therapeutic potential of IL-12/23 inhibition in animal models of colitis or other inflammatory conditions.
