Anti-p40 antibodies are a class of monoclonal antibodies that target the p40 subunit (IL-12B) shared by the heterodimeric cytokines Interleukin-12 (IL-12) and Interleukin-23 (IL-23). By neutralizing the p40 subunit, these agents inhibit the signaling of both IL-12 (which drives Th1 responses) and IL-23 (which drives Th17 responses), both of which are critical mediators in the pathogenesis of various immune-mediated inflammatory diseases. The most well-known member of this class is ustekinumab (Stelara), which is widely approved for the treatment of plaque psoriasis, psoriatic arthritis, Crohn's disease, and ulcerative colitis. In preclinical research, such as the IL-10 knockout mouse model of colitis, anti-p40 antibodies are frequently employed as benchmark controls to evaluate the efficacy of novel anti-inflammatory or barrier-restoring therapies.
