Overview

Anti-PAI-1 antibodies are monoclonal antibodies designed to target and neutralize plasminogen activator inhibitor-1 (PAI-1), a member of the serpin superfamily and a primary inhibitor of tissue-type plasminogen activator (tPA) and urokinase-type plasminogen activator (uPA). PAI-1 plays a significant role in regulating fibrinolysis and is implicated in various pathological processes, including airway remodeling, fibrosis, and type 2 inflammation. In respiratory models, PAI-1 has been shown to enhance the production of thymic stromal lymphopoietin (TSLP) in response to viral infections (such as rhinovirus) and allergic stimuli. By blocking PAI-1, these antibodies can reduce eosinophilic inflammation and potentially mitigate the severity of asthma and other allergic airway diseases. While primarily used as research reagents to study the PAI-1-TSLP pathway, anti-PAI-1 therapeutic candidates are being explored for their potential in treating cardiovascular, fibrotic, and inflammatory conditions.
